The most significant regions are probably Bordeaux, Burgundy, Loire Valley, and Côtes du Rhône, though there are plenty of others.  As for the biggest cities in/around each, I would say...
Bordeaux:  Bordeaux
Burgundy:  Dijon, Lyon
Loire Valley:  Nantes, Tours
Côtes du Rhône:  Avignon

I don't know that you'll find a Fodor's published book that will be much more useful than Fodor's France, which devotes about 50 pages to each of the regions.  If you're willing to venture away from Fodor's though, you might take a look at these two:
